Science, Ingredients & Compatibility

What are defensins?
Defensins are naturally occurring molecules found in the human body, best known for their role in the skin's wound healing process. Scientists discovered that every time a scratch or wound heals, the new skin that forms in the healed area always appears younger, firmer, and smoother than the skin surrounding it. This happens because the body calls upon a reserve of preserved, dormant cells known as LGR6+ master cells to generate fresh, new skin during healing. In 2014, researchers in regenerative medicine identified defensins as the key molecules responsible for activating this process. DefenAge's scientists harnessed this discovery to develop a patented topical formula that supports the same visible skin renewal response, without any wound required. Applied to intact skin twice daily, DefenAge's Age-Repair Defensins® work in partnership with the skin's own LGR6+ master cells, the same cells the body holds in reserve for renewal, to support the visible appearance of fresher, younger-looking skin over time. Unlike retinols, acids, or traditional anti-aging ingredients that work primarily by exfoliating or irritating the surface, defensins support the skin's natural renewal biology from within, making them a genuinely different approach to visible anti-aging. DefenAge's formula utilizes two specific defensin peptides, Alpha-Defensin 5 and Beta-Defensin 3, each targeting a different layer of the skin to support a complete visible renewal response.
How do DefenAge's defensins work in the skin?
DefenAge utilizes two specific defensin peptides, Alpha-Defensin 5 and Beta-Defensin 3, that work simultaneously in different layers of the skin to support a complete visible renewal response. Alpha-Defensin 5: Surface Renewal Alpha-Defensin 5 works in the epidermis by partnering with LGR6+ master cells, the same dormant cells the body activates during wound healing, to help generate fresh new skin cells that rise to the surface. The visible result is improved texture, radiance, and overall skin quality. Beta-Defensin 3: Deep Regeneration Beta-Defensin 3 works deeper in the dermis, supporting fibroblast activity to contribute to visible improvements in firmness, elasticity, and skin resilience. It also supports the skin's natural pigment regulation process, which is why many users notice improvements in uneven skin tone and age spots. Why Both Layers Matter Skincare products largely address only the skin's surface. DefenAge's two-peptide approach supports visible renewal in both the epidermis and the dermis simultaneously, producing results that are progressive and broad in scope. Both peptides are bioidentical, meaning they are identical to the defensin molecules your body already produces and recognizes, making them exceptionally well tolerated across all skin types.
What makes DefenAge different from retinol?
Retinol has been the gold standard in anti-aging skincare for decades, and for good reason. It works. But it comes with real trade-offs that many people struggle with: dryness, peeling, irritation, increased sun sensitivity, and the need to pause or reduce use in summer months. DefenAge takes a fundamentally different approach. Rather than pushing existing skin cells to turn over faster, DefenAge's Age-Repair Defensins® work in partnership with the skin's own LGR6+ master cells, a dormant reserve that the body holds for renewal. The visible results are similar, but without the irritation, sensitivity, or seasonal restrictions that come with retinol. This also makes DefenAge an excellent option for people who cannot tolerate retinol, including those with sensitive skin, rosacea, or those who are pregnant or breastfeeding. DefenAge and retinol are not mutually exclusive. Many dermatologists recommend using both together. If you use retinol or tretinoin, simply apply it before your DefenAge products as part of your routine. Does DefenAge contain stem cells, exosomes, or growth factors
No. DefenAge does not contain stem cells, exosomes, growth factors, or any human or animal-derived cells or tissues. This is a deliberate formulation decision. Stem cells, exosomes, and topical growth factors have attracted significant attention in prestige skincare, but they come with meaningful questions. Human and animal-derived ingredients carry potential infection and disease transfer risk. Topical growth factors are large molecules that struggle to penetrate the skin effectively, and some research raises questions about their long-term safety. Exosomes, while promising, remain largely undefined in terms of what specific proteins are active and why results vary between batches and sources. DefenAge's Age-Repair Defensins® take a different approach entirely. Rather than applying undefined biological material to the skin's surface, defensins are fully synthetic, bioidentical molecules, identical to those your body already produces, that work through defined receptor-mediated pathways to support visible skin renewal. The same result, with none of the variability or safety concerns. DefenAge uses biotechnology and plant-based ingredients exclusively, and every ingredient undergoes rigorous testing and quality controls before any product is released.
Is DefenAge clinically tested?
Yes, and extensively. DefenAge's Age-Repair Defensins® are backed by over 30 clinical and consumer studies, making the science behind the technology among the most rigorously validated in prestige skincare. The landmark study, a multi-center, double-blind, vehicle-controlled clinical trial published in the Journal of Drugs in Dermatology, evaluated DefenAge's three-step Clinical Power Trio regimen across multiple dermatological measures including histopathology, immunohistochemistry, ultrasound evaluation, and photographic analysis. Participants showed globally visible improvements in fine lines, wrinkles, pore size, pigmentation, skin texture, hydration, and overall skin quality within six weeks, with results continuing to improve through twelve weeks. Additional studies have evaluated individual products across specific concerns including skin thickness, eye area improvement, neck firmness, hair follicle density, and body skin quality. All studies were conducted without the irritation, inflammation, or sun sensitivity associated with retinoids. DefenAge does not conduct or commission animal testing of any kind. All safety testing is performed exclusively on human volunteers. Independent consumer studies further support the clinical findings, with 90% of participants reporting visible improvements in wrinkles and jawline firmness. Why do skincare professionals recommend DefenAge?
Dermatologists and plastic surgeons recommend DefenAge for a straightforward reason: their patients see real, visible results. DefenAge's Age-Repair Defensins® complement professional treatments including injectables, laser resurfacing, and microneedling, continuously building visible results between and after procedures. Many professionals describe it as a topical biostimulator, a non-invasive daily product that supports visible skin renewal from within. The clinical evidence carries weight too. The landmark Journal of Drugs in Dermatology study concluded that DefenAge globally improves the visible appearance of aging skin, offering the advantages of retinols and growth factor cosmeceuticals without irritation, inflammation, or sun sensitivity. Plastic surgeon Gregory Chernoff, MD, Indianapolis, notes that patients aged 18 to 90 show consistent improvements in skin tone, quality, and softening of lines. Dermatologist Natalie Curcio, MD, Nashville, adds that with DefenAge, skin can naturally turn back the clock.
Is DefenAge safe to use?
Yes. DefenAge is formulated to the highest safety standards in the industry and is suitable for all skin types, including sensitive skin. Every DefenAge formula undergoes a 30-Day Human Repeat Insult Patch Test (HRIPT), the gold standard for evaluating irritancy and allergy potential in leave-on skincare products, reviewed and signed by a board-certified dermatologist. Products used around the eye area undergo an additional 14-Day Ophthalmologic Safety Evaluation, reviewed by a board-certified ophthalmologist. DefenAge is also committed to clean beauty standards that go well beyond industry norms. All formulas are fragrance-free, vegan, and cruelty-free, and free from parabens, sulfates, phthalates, formaldehyde, hydroquinone, mineral oil, gluten, and chemical sunscreens. DefenAge does not use human or animal-derived ingredients of any kind, eliminating any infection or disease transfer risk associated with cell or tissue-based skincare. Unlike retinoids and many exfoliating acids, DefenAge's Age-Repair Defensins® support visible skin renewal without causing inflammation, dryness, or sun sensitivity, making it safe for uninterrupted, year-round daily use. For a full review of DefenAge's ingredient standards, visit our Clean Beauty page.
Is DefenAge fragrance-free?
Yes. Every DefenAge formula is completely fragrance-free. This is a deliberate formulation decision. Fragrance is one of the most common causes of skin sensitivity and irritation, and eliminating it entirely ensures DefenAge is suitable for even the most reactive skin types. It also makes DefenAge a safe choice for those with fragrance allergies or sensitivities. DefenAge also does not use scent-masking ingredients. Any natural odor you may notice is simply the natural scent of the active ingredients themselves, a sign of a genuinely clean formula with nothing hidden.
Does DefenAge contain sunscreen?
No. DefenAge products do not contain sunscreen or SPF. This is intentional. Combining active skincare with sunscreen can compromise the performance of both, and DefenAge's formulas are optimized to deliver the highest concentration of Age-Repair Defensins® without the dilution that comes with multi-tasking formulations. For daytime use, apply your preferred broad-spectrum SPF 30 or higher on top of the 8-in-1 BioSerum as the final step of your morning routine, followed by makeup if desired. This ensures both your DefenAge regimen and your sun protection are working at full efficacy. Unlike retinol, DefenAge does not increase sun sensitivity, so there are no seasonal restrictions on use and no need to adjust your routine in summer months.
Is DefenAge safe during pregnancy or breastfeeding?
DefenAge was designed with clean beauty in mind. All formulas are fragrance-free, vegan, and free from many of the ingredients most commonly flagged during pregnancy, including retinols, parabens, phthalates, hydroquinone, and chemical sunscreens. That said, DefenAge does not have formal clinical studies specifically evaluating safety during pregnancy or breastfeeding. Out of an abundance of caution, we recommend consulting your OB-GYN before beginning or continuing any new skincare regimen during this time. Your physician can review the full ingredient list for each product, which is available on the individual product pages on our website. For a complete overview of DefenAge's ingredient standards and excluded ingredients, visit our Clean Beauty page.
Can I use DefenAge with retinol or tretinoin?
Yes. DefenAge is fully compatible with both retinol and tretinoin and can be used as part of the same daily routine. Many dermatologists actively recommend using both together, as they work through different biological pathways and address complementary aspects of visible skin aging. The key is application order. Retinol or tretinoin should be applied before your DefenAge products, as part of Step 2 in your routine, after cleansing and before the 24/7 Barrier Balance Cream and 8-in-1 BioSerum. Applying DefenAge's Age-Repair Defensins® as the final skincare step ensures the defensins can work uninterrupted at the skin's surface overnight. One practical note: if you are new to combining both, consider introducing them gradually, particularly with prescription tretinoin, to allow your skin to adjust. DefenAge itself does not cause irritation, but tretinoin can be potent for some skin types and starting slowly is always advisable. Can DefenAge be used with Vitamin C?
Yes. DefenAge is fully compatible with Vitamin C serums and can be used together in the same routine. Vitamin C and DefenAge's Age-Repair Defensins® address different aspects of visible skin aging. Vitamin C focuses primarily on brightening, antioxidant protection, and collagen support, while defensins support the skin's broader visible renewal process. Used together, they complement each other well. For application order, apply your Vitamin C serum as part of Step 2 in your routine, after cleansing and before the 24/7 Barrier Balance Cream and 8-in-1 BioSerum. As always, the BioSerum goes on last.
Can DefenAge be used with cosmetic procedures?
Yes, and many aesthetic professionals consider DefenAge an essential part of their procedure protocols, both before and after treatment. 2 to 4 Weeks Before Your Procedure Starting DefenAge in advance helps optimize visible skin health before treatment, supporting skin resilience and overall condition going into the procedure. The First Week After Your Procedure For less invasive procedures, the 8-in-1 BioSerum can typically be reintroduced within the first week as tolerated. For more invasive treatments where the skin barrier needs additional support, DefenAge's 7X Calming Complex is an ideal first step, formulated specifically to soothe, calm, and support the skin's barrier during recovery. Two Weeks and Beyond Continue your full DefenAge regimen for extended visible support, helping to maintain and build on the results of your procedure over time. As always, follow your provider's specific post-procedure instructions, and consult with them before resuming any skincare products after treatment.
